Could be DVT: If it is truly calf and not ankle pain it could be a dvt. You should seek care and get an examination. The colonoscopy might be totally unrelated. Other considerations include tendinitis or a muscle strain.

Possible DVT: Calf pain with DVT usually exhibits some swelling. You were immobile at times and could have developed stasis. See doctor for exam, ultrasound to find out without delay.

Calf pain: certainly this calf pain is of concern, and may represent a DVT but there could be other possible causes. The first evaluation should be endovenous Doppler sonogram which would definitively answer the question if there is a deep vein thrombosis or not. If not further evaluation and perhaps by orthopedics will be of benefit.
