No single one: There is no one single "best" weight loss exercise routine--"best" needs to be tailored to the individual's capacities, limitations, schedule, etc. Weight loss pills are in general a trap as weight lost while using them tends to rebound ferociously upon discontinuation--and they shouldn't be used permanently. For life-threatening obesity the answeris more complex.

Pills...NOT advised: The best routines are those that allow motivated individuals to succeed at achieving their specific goals. Running will improve aerobic fitness but won't do much to increase muscle mass. Sprinting will increase endurance but may not increase aerobic capacity. Being specific will help identify the right programs you need.
