To know the effective (actual) dose level of any medication or supplement you get a sample after any new dose has been widely distributed in the body. These materials take time & may need to bind to carrier proteins to be effective. Samples taken too soon may give faulty results. The cited levels are just markers for comparison at different intervals.

Some of the testing methods use biotin conjugated reagents and high levels of biotin in the serum/plasma can interfere with the testing method and provide erroneously high or low results.
