Having muscle spasm with pain and soreness in the lower back and upper thigh muscles, gets worse with sitting, so what is the fastest and best trestment, also tired the conventional topical and systemic therapy with not that great relief for week ?

check your posture and correct it if needed. try local heat and physiotherapy. if not better it needs investigating by your MD with X-Rays etc.
