Is taking diclofenac 75 mg twice a day more effective than taking diclofenac 100 mg er 24 hr dose?

May be: Some patients get better results taking this medication twice a day. You would have to try both doses to see which works better for you. This is an anti-inflammatory medication and not a straight pain medication(like Tylenol or opiods) although it is used as a pain reliever.
