Great question!: Vyvanse, (lisdexamfetamine) like all stimulants, should work the first day you take an adequate dose. It differs from other stimulants in last longer; perhaps in your case, there is a bit of build-up, so you "right dose" doesn't happen until a day or so after you start. If you've used other stimulants and had a similar response, though, that would not be the explanation.
