Depends: Depends on the individual. I think it is well accepted that it is usually 2 weeks before Amitriptyline reaches maximum effect in the human body.

A lot: Although Elavil (amitriptyline) is an antidepressant, the medicine is out of traditional repertoire for depression. Very high dose will be required, and may be poorly tolerated.
