No: The material that the line is on wicks the fluid along it. Tilting it won't make any difference in the result.

Depends on the brand: Some brands and pill styles are more likely than others to give a false negative (says you are not pregnant when you really are) with manipulation. While i suspect the negative is accurate- the water usually gets pulled across the line independent of gravity. It would be reasonable to re-check the test in a couple of days without tilting it.
