Maybe: Only you and your partner can determine when is the best time to concieve. Ideally, you should be up to date on your health screening (pap smear, immunizations), you should be taking a prenatal vitamin and i recommend preconceptional counseling visit with your ob/gyn prior to conception.

If you're trying : Have yourself in proper physical, nutritional, emotional and financial health, if at all possible. Be aware of family genetics on both sides. Do appropriate screening for inherited disorders. No alcohol, no tobacco, no marijuana, no drugs in a woman of child bearing age who intends on becoming pregnant.

Many methods.: The week around ovulation is the best time to get pregnant. Having sex daily or every other day during that week gives the same preg rate. If you have regular cycles, 14 days before your cycle should start is the most likely ovulation day. If you can do an ovulation calendar with basal temp check and cervical mucus, that is even more accurate. Ovulation testing kits (available otc) are even better.

Ovulation: Shortly before/around ovulation, which is usually mid-cycle. The home opks are pretty reliable if you choose to use one; you can also subtract 14-16 days from your average cycle length and you'll be pretty close.
