Depends: Currently, surgical excision is the standard of care. However, depending on the stage of a patient's melanoma there are some new treatments that may show promise for the future. Vemurafenib is one such treatment, but it is used in a very specific set of patients. Melanoma vaccines are also theoretically possible but have not been very successful to date.

Depends on situation: It is very exciting that three (yervoy, zelboraf, and sylatron), and possibly a few other, new drugs have been approved for advanced melanoma treatment since the beginning of 2011. The best? That's a very variable question because none of the drugs are perfect yet all of them are, in the correct situations and managed appropriately, potentially helpful.
