Not necessary: Unless told told to do so by your doctor. They are designed for weight bearing so unless your a sleep walker not necessary.

Talk to your Dr.: Often times it depends on what is being treated and what the doctor is trying to achieve with the splint. If sleeping with it, make sure it is not tight and cutting off blood circulation.
