Xray and biopsy: Infants suspected of hirschsprung's will need a barium enema or lower GI series to study the large intestine. If suspicious then a biopsy of the lining of the rectum is required to assess whether ganglion cells are present. If the nerve cells are absent then one is diagnosed with hirschsprung's disease and additional surgery can be undertaken to cure the patient.

Usually enema/biopsy: Usual test for Hirschsprung disease is barium enema. If pattern on enema suspicious for disease, biopsies of rectum and colon performed looking for presence or absence of ganglion cells.Rectal manometry is used by some gastroenterologists. Pressure differences between ganglionic and aganglionic segments.
