Several: Mostly depends on the kind of problem, but in general urine analysis, sperm analysis, ultra sound of the prostate, biopsy, sometime giving a med can be a test as well as a potential help. Please understand these tests are only done when indicated!

In blood or urine...: If you refer to blood tests we usually order psa in adults above age 50 ( which has been controversial )or before if symptomatic or if the dre(digital rectal examination) shows nodules, or enlarged or painful prostate. The urine analysis may be positive for infection or blood in cases of prostatitis. The pap ( prost. Acid phosphatase) is ordered in cases suspicious of prostate cancer. Others.
