A single daily dose: of Vyvanse (lisdexamfetamine) has an onset of 1 1/2- 2 hours, peaks around 6 hours, wears off at 12-14 hours, & stays in your urine for 2-3 days. With prolonged use, amphetamine can be detected for 3-5 days after the last dose.
