Cortisone, yes: Hydrocortisone and similar steroids are indeed helpful in alleviating the inflammation and swelling around a hemorrhoid. Nystatin is an antifungal and unlikely to be helpful. You may want to check to be sure your steroid is of a strength that's safe to use in such a delicate area.
