Tightness: If you are experiencing tightness, this is more likely your facial nerve.  Also, could be something associated with your electrolytes. CN 5 should move your jaw, CN 7 moves rest of your face.

Many factors: The pathophysiology of headaches involves many things, from releases of certain neurotransmitters to changes in blood flow and permeability of vasculature.  Subtle swelling on one side of the face or the other, and/or constriction of swelling, can give a differential subjective feeling.  Thus, one side of the head/face or other may be felt to be different.

If new: it means you were exposed to something (sun, chemical) to which you are allergic more on one side than the other.  You can also affect the puffiness of your face by sleeping on it wrong.  To stay symmetric, sleep with your head straight up towards ceiling (so lying on your back)
