Possible herpes: A blister like that is more concerning for herpes than HPV. I would have your doc check it out in office at first available appointment to see what it truly is and appropriate treatment for it. Primary genital herpes may require antiviral medication and warrants a trip to the doctor.  Avoid unprotected sex until it is properly diagnosed (condom mandatory or abstain)

Possible: But unlikely. There are very few illnesses or conditions that create a vesicular eruption (blisters). Occasionally this can occur through a contact dermatitis, but most commonly is herpetic in the area in question.

Blistering: Sexual intercourse most commonly causes redness or slight excoriations which may be uncomfortable.  A topical lubricant sometimes will help alleviate this.  Blistering is not typical and is most commonly associated herpetic infections even if your only symptom is an itch.

Blisters : Blisters on the vagina are worrisome. You could be dealing with herpes. Herpes can be contracted in other ways beside sexual contact, so this still needs to be ruled out. As far as the raised "brownish dot" this could simply be a common mole aka nevus. It is difficult to say without actually examining you myself.
