Does coughing after running always mean asthma?

No: Not necessarily, but asthma needs to be considered, so get to a doc to rule out.

Probably: Coughing that is worse with exercise, laughing, or crying or that wakes you in the middle of the night leans toward asthma. You can do a breathing test. If it is normal you can try an inhaler prior to running. Alternatively you can do a an exercise test where you do a breathing test, then exercise, then repeat the breathing test and look for a drop.. There is also a methacholine challenge.
