Is buscopan supposed to alleviate stomach pain after each dose or is it something that you would need to be taking for a few days before improvement?

Buscopan: Well, while it may work after each dose, it may also alleviate the overall condition as your bowels start being "retrained", so the cramps may feel less frequent and painful after an extended use of it.
