Unlikely: Enlarged lymph nodes are from inflammation or similar process. Masturbation alone will not cause this problem. Have your doctor investigate more thoroughly. Best wishes.

Indirectly: Any skin tear or abrasion in or around the penis could cause an enlarged lymph node in that area. I would recommend that you keep an eye on it and let your doctor know of any changes- such as enlargement.
