Varies: Most new generation antihistamines (allergy pills) such as allegra, zyrtec, claritin, etc can be taken once or twice a day meaning their good effects should last 12 to 24 hours. Older antihistamines like Benadryl (diphenhydramine) are taken up to 4 times a day meaning the good effect may last only 4 to 6 hours (and make you sleepy!) an allergist can assist in finding the right treatment plan.
