It's possible, but: Tattoo ink is deposited in the dermis, second-down layer of the skin. There it is internalized by macrophages, clean up cells of the skin. Because macrophages don't really know what to do with tattoo pigment, they just sit in the dermis holding on to it. That's sort of what keeps the ink in place. In a scar there aren't that many macrophages, so you may get unreliable results.
