My wife get blood spotting on her menstrual cycle only for two days that she never get period only for two days its her first time. but she feel that she is pregnant we checked with pregnancy test kit its showing negative. what to do next thanks?

Another test in 2-3 days with an early morning urine sample should be done. If test is still negative then chance of pregnancy is very less likely. It could be the first time she has an atypical period then just observe. Good luck.
