A semen culture involves material derived from the prostate gland, separate from the bladder or urinary system. The prostate can be infected and a urine culture not provide a valid sample to test for it.

Semen culture is usually done to check for infection of prostate or seminal vesicle( a leaf shape gland which stores sperms). Urine is the waste water we get rid of via the kidney/ bladder. So urine culture is done to find type of infection in that system
