Tramadol: I’d be cautious about combining these as there is a moderate risk of interactions. Best to avoid the 5 HTP if possible while on the tramadol or lower the dose and yes, space them out.

Not recommended: Taking 5-HTP and tramadol together may increase the risk of serious side effects, such as something called serotonin syndrome. It is important for your doctor to know you are taking these two medications and should be carefully monitored. If the two of you decide that you should take 5 HTP, then the doctor may prefer to prescribe a different pain medicine.
