If its Depo-Provera: You need to identify what "shot" you are talking about. A guess would be Depo-Provera [Depot medroxyprogesterone acetate (DMPA)] for birth control or other gynecologic problem which does have to be repeated every 12 weeks. Penicillin wont affect the action of Depo-Provera but it's possible whatever you were treating with the penicillin had some effect. You had your period cause the drug wore off.
