Within few minutes : An intramuscular shot of penicillin statrs to work within few minutes as soon as it is absorped in blood stream and keeps working till another shot is given for short acting procaine penicillin, but long acting penicillin is given as one shot for certain treatmemts and do not need repeat shots.
