ED or infertility?: Please clarify whether you're asking about erectile dysfunction (ED) or infertility (inability to father children). If former, it'll be obvious that he can't attain/maintain erection satisfactory for sexual activity. If latter, will need semen analysis looking for viable sperm.

Lack of erection: Impotence is lack of erection and does not need any tests. If you mean infertile, it should start with a physical examination, sperm count and sperm morphology.

Impotent : There are many factors that contribute to impotence. First is determine that your are impotent. Simple check is for early morning erections or erections with pornography or with a non partner. If those are present not impotent. May need sex counseling with partner or excessive weight, inactivity, diabetic and side effects of many medications like Beta blockers and anti depressants
