Imitrex (sumatriptan) at onset: Migraine medications are more effective when taken as soon as possible when the headache starts when the pain is mild so the relief may be less if you wait until the pain is moderate to severe. The maximum relief with each dose may not be felt until 4 hours so some people prefer waiting until after 4 hours until taking the second dose. If Imitrex (sumatriptan) does not work, another triptan might be better.

Imitrex (sumatriptan) after 2 hour: According to the fda-approved directions, it is recommended to take a second dose of Imitrex (the same with another triptan - maxalt, relpax, (eletriptan) etc) 2 hours after the first dose. The maximum dose for 24 hours is 200 mg, although the drug is washed out of the body in 4 hours and taking a third 100 mg dose within 24 hours is not dangerous, but each person is different, so check with your doctor first.
