A penis in vagina: Technically, if you have had a penis in your vagina, then you are not a virgin. Rape might be considered an exception. Some people would consider any form of sex such as oral sex or manual stimulation using a partners hand would qualify as not a virgin. The real question...What does being a virgin mean to you? Or your partner?

Hard to be specific: Most would consider any partner sex that involves the genitals would cancel this label. Some try to use an intact hymen as the marker, but some women are born without or lose it in non sexual events. Why someone would ignore anal or oral genital contact & still use the term is hard to understand.If you engage in a sexual contact that could pass a STD, it is hard to defend the "virgin" label.
