Several: An rh-negative pregnant woman is screened for antibodies against d (the main rh antigen) and pregnancy is followed for signs of trouble. If a previous or another type of mismatch is suspected, additional labs are done. If suspected in a newborn, baby gets a direct Coombs for antibodies on red cells.

See below: The cord blood can be tested in mothers who are type o and who's fathers are type a or b. Alternatively the blood can be tested on any newborn who is jaundiced.
