The only active ingredient in Prolia is denosumab, a monoclonal antibody that binds a signaling molecule, RANK Ligand (RANKL), that bone forming cells (osteoblasts) secrete that activates bone resorbing cells (osteoclasts). RANKL activates the osteoclasts to break down bone. Denosumab interrupts that, and preserves bone mass and architecture.
