Inflammation: If the pain is really in the ankle this could be an inflammatory or arthritic process. Many time this can be confused with heel pain or plantar fasciitis.

Various reasons: "start up pain" is often indicative of arthritis. Unless you have a history of injury to your ankle, this would be less likely in a 34 y/o. See an orthopaedic foot and ankle specialist if your pain persists.
