Urine concentration: Urine test can be just as positive at night as in the morning. Morning urine is usually more concentrated and therefore more sensitive. It is more important to do the test 4-5 days after a missed period.

Depends: Home pregnancy test should be done 4-5 days after a missed period. It is better to use first morning urine as it is more concentrated. However, if the blood HCG level is high enough you could do the test any time.
