No: Bactrim (sulfamethoxazole and trimethoprim) shouldn't negatively affect(weaken) your immune system but can cause antibiotic induced diarrhea by altering the bacterial makeup of your intestinal tract, but in my experience this side effect is much more common when other classes of antibiotics are used.

Not particularly: Any antibiotic you take will kill off any germ that is sensitive to it. That is in the gut or in your infection. Simply taking a probiotic or eating probiotic yoghurt will replenish what are killed. It has no other effect on your immune system.
