See below: The earliest time to take a pregnancy test would be about two weeks after sex. If the result is negative you should repeat the test in 4-5 days. Repeat use of Plan B is not likely to reduce its effectiveness; however if you do not wish to be pregnant use contraception consistently and talk to your doctor about regular birth control pills.
