Ejaculation: One ejaculation never empties the prostate gland. Semen and sperm are being produced round the clock and the job of these glands and organs are to keep the prostate gland full and ready at all times.

Vas deferens is duct: Sperm is produced in the testis, they pass into the vas (1) via the epididymis ; thence to the ejaculatory duct. Most of your semen is produced in the prostate gland (3) ; seminal vesicles (2). Some men are able to ejaculate a second time 15 - 30 minutes after the first. Men tend to produce less semen as they age ; thus take longer before they can ejaculate again. Hope this answers you.

In 1 day or less...: Are you conducting a study? Within less than one day, "normal" amount of ejaculate will restore, and in 3-4 days, it will return its "maximal" volume. Remember: the volume may vary individually and don't use this to gauge the timing and possibility to impregnate sex partners. This is based on my observation from 34-year medical practice.
