Yes: If you can, yes. The injury that caused the thumb to need a removable cast may be made worse by some thumb positions. Making sure that your thumb maintains the neutral positions during sleep is beneficial in most cases. However, if it really bothers you and you can't sleep wearing it, then sleep trumps the thumb.
