If you were to only dose the tobramycin once a day, it will be too low to work in ~12-16 hrs; by using it twice a day, you keep it in your system & it works much better.

Should be OK: These two medications are compatible with each other, but they are used for different purposes.  If you have an eye infection requiring the use of tobramycin, the treating doctor might want you to temporarily stop Restasis until the infection is resolved. Otherwise, there should no problem with using both.
