If you want to rule: Out a blood clot you do a venous duplex which will show you if a clot is present. I am not sure what blood test can do that....

Baker`s cyst: Baker`s cyst will not cause swelling in the leg. The cause of ankle swelling is probably venous insufficiency which is diagnosed with doppler and ultrasound studies. Baker`s cyst discomfort might persist, in which  ace you might consider surgery.

: You may have Athlete's foot which can cause redness, flakiness, peeling, or cracking of the skin on the feet. It sometimes has an itch, sting, or burn, or simply feel uncomfortable. It is located on the soles of the feet, the areas between the toes, and sometimes the toenails. You need to use an antifungal product such as clotrimazole. The swollen legs that hurt are aothe problem.

See a physician: It is important to know why you are having swelling; whether it the result of a local problem, such as an injury or a systemic cause. Also, symptoms and other local signs are also important. In the case of venous insufficiency compression stockings are a first line treatment. Conditions such as lymphedema require other approaches. See your physician for evaluation and appropriate referral.

Complex question: If the cause is poor venous circulation- elevation of legs frequently, compression hose, salt restriction and and leg toning exercise are all easy to do.

No: you have venous stasis.  Any other swelling can be like abscess formation, muscle herniation.  Fracture, and if you have the above trust me you will know it right away.   My advice if you have good arterial flow get 30-40mmHg compression sock and use them. Cut salt from diet and elevate feet when at rest
