Blood test is more: Sensitive than urine pregnancy test. It turns positive sooner if a woman is pregnant. Not that much more accurate but provides an answer sooner.

Yes: An ovarian cyst is most likely the reason for no menses now.If it is large enough to compromise the bladder capacity it would cause more frequent urination

May help: If you are 4-5 days past the day you were due for your period, it would be useful to do an hpt using first morning urine and following the instruction for the test carefully.

Maybe. Maybe not.: Any person having sex can catch a sexually transmitted infection. That's just the nature of STI's. Of course, the other person or one of the other persons have to be carrying an STI in order to transmit it. You can't get something from nothing. STI's do not typically cause a "lumpy vagina." A person with a "lumpy vagina" needs an in-person exam so her doctor can see what "lumpy" actually means.

Well, among other: Things i think u have a bladder infection.  It would explain a lot of the cramping.  The breast soreness is related to changing hormones.  You had a buffer with the depo-provera because your ovaries were not producing estrogen.  The leg pain may be referred from the bladder or uterus but if u have a swollen leg and difficulty walking it could be a blood clot. That's unlikely but i would see doc.

See the doctor: With persistent bloody urine, you really need to get it checked out. It could be something simple like an infection, but it could indicate other more serious things. Irregular menstrual bleeding is common with depoprovera, but usually after 9-12 months, bleeding stops. If you are beyond this time, and it is just starting to bleed more, ask your doctor.

Yes: If you were testing specifically for progesterone, then likely the test would show up in blood (i'm not sure about urine, I have never heard of a urine Progesterone test).  Women's ovaries also make progesterone, and those levels would also be accounted for.  It's an interesting question you pose, maybe consider re-posting with additional information as to why you're concerned/interested in result.
