It's more complicate: When a neuron fires, after it does so it will reabsorb or "reuptake" some of the serotonin it released when it fired. To inhibit this process, the neuron needs to change so it has fewer openings available at that end. The drug causes some of the openings to close, a change which takes a while. It's called "down-regulating."

Not exactly,...: Latuda (lurasidone) treats schizophrenia and bipolar depression, but its exact actions inside the brain is unknown. It may work by blocking certain dopamine and serotonin receptors in the brain. That means it prevents dopamine and serotonin from attaching to those receptors, which are locations on brain cells that dopamine and serotonin would attach to if Latuda (lurasidone) molecules were not blocking those locations.

Medical vs psycholog: There are both mind and body causes of unhappiness and hopelessness.  Low serotonin, hypothyroidism, and other medical conditions can impact mood.  Psychologically things like loss, significant change (positive or negative) or other factors can impact mood in a negative way.

Depression: I don't know how long you've been off the zoloft (sertraline), (sertraline) but lost of interest and pleasure is more consistent with depression than with zoloft (sertraline).  If your depression symptoms are rising again, you need to see your psychiatrist to review treatment options -- possibly a different med, depending on evaluation.  If this is from depression, psychotherapy may be helpful to you also.

: If your SSRI has been taken for a few weeks &amp; hasn't been effective, it's worthwhile switching to a different SSRI as efficacy can be highly individual with these meds.  However,the transition must be done carefully &amp; must be supervised by your prescribing physician in order to minimize withdrawal problems, side effects or aggravating your original symptoms.

Stop Supplement: I agree that stopping the supplement is a good idea.  I would wait and repeat test a few weeks after that, unless you are experiencing flushing or other symptoms of serotonin syndrome in which case your doctor should do a work-up for carcinoid or other serotonin secreting rumors.
