Its a progesterone: it is generally used to keep cycles going as one part of birth control pill regimen. Generally it is used after a woman takes estrogen. You take duphaston, then stop everything and get a period.....so... in this context, yes.

: If you had sex since your last period and missed your period by 4-5 days, do a home pregnancy test. If the result is negative repeat the test in 4-5 days, unless you get a period. Use first morning urine and follow instructions for the test carefully. If you do not wish to be pregnant, use contraception all the time, every time. You may consider implanted contraceptive, or IUD. 
Get HPV vaccine.

Predicting Ovulation: You can monitor your ovulation using Ovulation predictior kits (OPKs) sold in major drug stores. Talk with your GYN to help you determine when to start and for how many days given irregular periods with PCOS. OPKs help detect the LH surge, which occurs 12 to 36 hours before ovulation. Just follow the instructions that come with the kit. If that fails, talk to your GYN about other methods like US.

Duphaston: If withdrawal bleeding took place on day four of Duphaston, there is no need to continue if the reason you were prescribed Duphaston was that your period was delayed.  This is an "artificial" period, and if fertility or other menstrual issues continue, see your ob/gyn provider for an exam and other testing.

Soon: Usually the period will come 2 or 3 days after the medication is stopped, so give it a little more time.  Sometimes it will not bring a period, but take it a step at a time.

Clarify with doctor: Clarify the doctor who prescribed the medication how you are supposed to be taking it. Progesterone is sometimes used to regulate cycles but one would not expect a cycle until you stop taking it.

Progesterone: Progesterone does not always work to trigger a period in all patients. It is not clear why you are taking this medication. If you think you might be pregnant check then check a home pregnancy test. They are cheap, reliable and available  at any pharmacy. Follow up with the prescribing doctor to discuss why you were given this and what other options may be available to meet your goals.
