Tests are excellent: If done properly they are very good.  Your HCG levels should be at least 20, 000. The tests can detect HCG levels about 1000 times less than that.

After missed period: You may take the test any time you wish, but it to be better to wait 4-5 days after a missed period. Use first morning urine and follow the instructions for the test carefully.

At least 2 weeks: Conception may take  place only if  sexual intercourse happens within few days from  ovulation. After conception it takes about 2 weeks to produce detectable levels of HCG, the hormone responsible for a positive test.

Depo contraception: Depoprovera injections typically are very successful at pregnancy prevention though not 100% fool proof.  The longer on depo, the longer it will take you to ovulate and then become pregnant. If you are trying for pregnancy you should be off the depoprovera and your ovulation pattern will start again in 3-6 months from the last Depo-Provera shot.

Yes: If the last episode of sex was more than three weeks ago, had you become pregnant hpt would be positive. Use first morning urine and follow the instructions for the test carefully. If the result is negative and you do not get your period, you may wish to re do the test in 4-5 days.

Pregnant: By hpt I assume you mean home pregnancy test?  If you have missed a period would check the test again. If you have not, you are not pregnant. The doxycycline is unlikely to have influenced the efficacy of Plan B. You might want to discuss other means of contraception with your gynecologist. Good luck.

After missed period: You may get a blood test one day after a missed period. Urine pregnancy test may be done 5-7 days after missed period or three weeks after sex. If the result is negative, repeat the test in 4-5 days. Use first morning urine and follow the instructions for the test carefully.
If you do not wish to be pregnant, use contraception all the time, every time. You may consider implanted contraceptive, or IUD. 
Practice safe sex.

Repeat pg test: Repeat the pregnancy test in a week after a missed period.  If negative and no period, consider seeing your doc for a blood pregnancy test.  Also, see the doc to get checked for sexually transmitted diseases if you have not been in a mutually monogamous relationship for a year.

No: Conception, as defined by the generation of a fertilized ovum, occurs within day or so of the sex. However, it takes about two weeks for the embryo to make enough HCG to produce a positive pregnancy test. Blood test may be done a day after a missed period. Home pregnancy tests should be done 4-5 days after the missed period. Use first morning urine and follow instructions for the test carefully.
