I assume the : Blister was drained? So, the question is is there raw skin that is hurting? Is it possible you developed an infection? Is there a bandage in or did you take it off. If you took it off what do you see?

Leave it alone: Blisters are your bodies defense to repair the damage, don't pop it. Keep it clean and dry. You may want to fix the cause ie.Shoes too tight or too loose. (friction).

See a doctor!: (only because you said 'big'). You can try popping it with a sterile (not hot) pin on the edge, just where the blister meets normal skin and then wash with soap & water and keep covered with a sterile band aid. Watch for signs of infection (redness, pain, pus, odor, swelling) and if you think it's infected, see your doctor.

I peeled off: See a podiatrist  for PO antibiotic and wound healing products such as amerigel wound dressing with topical antibiotic/ fibracol collagen dressing that work very well and quickly heal the wound. If you have diabetis, do not wait, the infection can spread quickly.

Pressure: If you have already opened the blister (i assume it is an irritation blister), then i think you should press hard and remove all the liquid (serum) from the blister, so that the covering skin lays flat, it should act as a matrix to heal that way. If concerned about infection you can spread a bit od neosporin, Bacitracin or polysporin/triple antibiotic on it.

Itch by the inch.: It sounds to me like you are experiencing the discomfort from a fungal infection. It might be best to start treatment soon as it can spread quickly. Try over the counter medications first. If there is no improvement, then see a specialist.
