First void is best.: The time of day you take a pregnancy test does matter. Home pregnancy tests work by detecting the hormone human chorionic gonadotropin (hCG) which is more concentrated in your urine when you first wake up. You can still take a pregnancy test in the middle of the day, or even at night, but these times of day are more likely to give you a false-negative result.

Do the test: If you had sex since your last period and missed your period by 4-5 days, do the test. If the result is negative, repeat the test in 4-5 days, unless you get a period by then. You may get a blood test for HCG level to settle the issue now. Wish you good health!
