Once : Once a patient has had mono, they will always test positive for the virus. In general, patients only get true mono one time, but if the patient is very "run-down", or susceptible from physical stressors, the virus can re-activate and cause mono symptoms and can also be contagious again.

No, but....: The classical mono is caused by EB virus, and you only get it once. Afterwards you will have lifelong immunity. However, there are other germs that can cause a mono like illness. Acquisition of this 2nd or 3rd germs could give you a similar illness.
