Chromosomal analysis: Kleinfelter syndrome is caused by an extra x chromosome in males (47, xxy). It can be suspected on physical exam because of several somatic features including small testis in post pubertal males. Definitive diagnosis requires chromosomal analysis.

Genetic testing: Although you may have low testosterone levels or elevated levels of LH and/or FSH (which are the signals from your pituitary gland which stimulate testosterone production) the most accurate test is a karyotype, which will usually show a 47,XXY pattern, which means you have two X chromosomes and one Y chromosome, if you have Klinefelter's.
