Both test Ag-Ab rxns: Direct Coombs will test for the reaction using the patient’s RBCs, if there is a reaction, then the direct test is positive, used to test for autoimmune hemolytic anemia. Indirect Coombs test uses the patient’s serum (not rbcs) which is added to rbcs that are known to have a certain antigenicity, positive in cases of hemolytic diseases of the newborn, mother's serum against baby's RBCs,
