No: amniocentesis is the process of obtaining fluid from the gestational sac, passing the thin needle under continuous ultrasound guidance through the uterine wall. It is no more dangerous to uterine muscle than an injection would be to a gluteal muscle. After a Cesarian, the uterine wall does just fine after bering sewn up with multiple stitches.
